I've only used modern materials for heavy bow strings. I've heard of people experimenting with hemp. I'd suggest using the same formula as for any other bow.
I'd be more concerned about wear at the nocks than the actual string count

It would have to depend on the quality of the linen you have.  You would need to find out what break strength the linen has then figure out the number of strands to at least 4 times the weight of the bow.  Even then if the quality of the linen is poor then it might not be enough.  I have some linen here that I wouldn't hesitate to make a string for a heavy weight bow, heaviest bow I have used the linen on is 65#.  If you are using the pyro linen then I wouldn't bother
